Peptide Storage Guidelines

Peptides are biologically active molecules sensitive to temperature, moisture, light and repeated freeze‑thaw cycles. Proper storage is critical to retain peptide activity, prevent degradation and extend shelf‑life. Below are standardized storage recommendations for lyophilized peptides and peptide solutions.

Lyophilized (Powder) Peptides

Lyophilized peptide powder is the most stable form.
  • Long‑term storage (More than 6 months): Store at ‑20 °C or below, under dry and dark conditions. Seal tightly to avoid moisture intrusion. Under this condition, most lyophilized peptides can remain stable for 1‑2 years.
  • Short‑term storage (1‑3 months): Store at 2‑8 °C (refrigerator). Avoid long‑term placement at room temperature.
  • Important note: Do not open the vial immediately after removing it from cold storage. Allow the vial to warm to room temperature before opening. This prevents condensed moisture from entering the vial and causing peptide degradation.

Reconstituted Peptide Solutions

Once lyophilized powder is dissolved into liquid, peptide stability decreases significantly.
  • Long‑term storage of peptide solution: Aliquot into small centrifuge tubes and store at ‑20 °C or lower. Avoid repeated freeze‑thaw cycles, as each cycle will accelerate peptide decomposition.
  • Short‑term use: Store at 2‑8 °C refrigeration and use within 1‑2 weeks.
  • Never store prepared peptide solutions at room temperature. Aqueous peptides are susceptible to hydrolysis, oxidation and bacterial contamination.

Special Reminders for Specific Peptides

  1. Peptides containing cysteine (Cys) or methionine (Met): Prone to oxidation. Minimize air exposure; inert‑gas sealing is recommended when available.
  2. Readily hydrolyzed peptides: Avoid repeated dissolution and freezing. Prepare working solutions immediately before experiments.
  3. Light‑sensitive peptides: Keep away from light. Brown storage vials are preferred.

Common Storage Mistakes To Avoid

❌ Opening cold vials directly without temperature equilibration, which causes moisture condensation
 
❌ Subjecting peptide stock solutions to repeated freeze‑thaw cycles
 
❌ Keeping peptide solutions at room temperature for multiple days
 
❌ Exposing peptides to strong light or humid surroundings
